U-143 sails on her first patrol cruise from Kiel to the south coast of Iceland, Kiel, Germany, 19.04.1941.

The first two weeks of the voyage were unsuccessful. It was not until 03.05.1941 at 20.50 that a patrol spotted the convoy and the submarine's captain Harald Gelhaus decided to follow it at a safe distance. The Submarine Fleet Commander-in-Chief Dönitz received the news of the sighting of the convoy in the area with great scepticism and asked Captain Gelhaus to confirm it. Only after the report was repeatedly confirmed did Dönitz order the submarines U-141 and U-147 to move to that area. At 0100 the following day, U-143 reported to HQ that she had lost visual contact with the convoy. The convoy was not encountered by the two submarines dispatched. The reconnaissance aircraft dispatched reported sighting only a few ships sailing independently. After 25 days at sea, U-143 returned to port Bergen, Norway on 13 May 1941.
